Mandate field Entry based on another cell value

Hi All,

Need little help in writing a VB code for the above criteria.
I am trying to mandate few fields of respective columns entry in a sheet.
For example if cell AF3 contains text value then cell O3 should ask for mandatory entry.

Secondly, suppose if the cell M3 contains some value (number) then it should equal to the sum of cell N3, O3, and P3 and if the value doesn't match then it should ask us to correct it.

This requirement should doesn't applicable particularly to one row but it should active rows.
I mean this rule applicable only when the first column say "A" has some serial no. entered in it.
